The paradigm shift

Shifting the scaling problem from chips to compute.

Digital PCR provides absolute nucleic acid quantitation with exceptional precision. Conventional systems achieve it through complex and expensive microfluidics. DPlexBio takes a simpler path.

Conventional digital PCR

Precision is built into the consumable.

Microfluidic chips create highly uniform partitions before PCR.

Scaling depends on precision-manufactured physical components.

DPlexBio edPCR

Precision is built into the measurement.

Simple shaking creates variable-volume droplets that are measured by 3D imaging and AI analysis.

Scaling shifts toward imaging, software and computation.

edPCR replaces microfluidic partitioning with 3D imaging and computation.

Simple workflow

Digital PCR, simplified.

Generate droplets directly in standard-format wells, thermocycle, image in 3D, and let computational analysis determine absolute target concentration.

A different approach

Different-sized droplets are not a limitation. Polydispersity is an advantage.

Conventional digital PCR has been built around the assumption that accurate quantitation requires uniform partitions. We have demonstrated that accurate droplet digital PCR does not require monodisperse droplets—and, in fact, benefits from polydisperse droplets — and thus does not require an expensive instrument or microfluidic chip to generate them.

Simple droplets. Sophisticated measurement.

Computational measurement

From fluorescence images to molecular counts.

DPlexBio’s emulsion digital PCR (edPCR) software identifies individual droplets in 3D, measures their volumes and fluorescence, classifies PCR-positive reactions, and calculates absolute nucleic acid concentration.
